Characterisation of cauliflower mosaic virus DNA sequences which encode major polyadenylated transcripts.

چکیده
Cauliflower mosaic virus DNA sequences which encode two major polyadenylated RNA species, the 1.9 kb messenger RNA for the 62,000 MW virus inclusion body polypeptide and 35S RNA, were mapped using the nuclease 51 procedure. The 1.9 kb RNA has an eleven nucleotide leader sequence transcribed from alpha-strand DNA located at co-ordinate 0.72 m.u. (map units), immediately upstream of the AUG initiation codon of reading frame VI. The 3'-end of 1.9 kb RNA maps at 0.95 m.u. and is co-terminal with the 3'-end of 35S RNA, a complete transcript of the DNA alpha-strand. The 5'-end of 35S RNA maps at 0.93 m.u. and is located upstream of the discontinuity G1 (zero m.u.) in the alpha-strand and some 200 nucleotides upstream of the sequence encoding its own 3'-terminus.

Production of aberrant promoter transcripts contributes to methylation and silencing of unlinked homologous promoters in trans.
Previous work has suggested that de novo methylation of plant nuclear genes can be triggered by an RNA-DNA interaction. To test whether transcription of a promoter would induce de novo methylation and silencing of unlinked genes driven by the same promoter, a chimeric 'gene' consisting of a nopaline synthase promoter (NOSpro) positioned downstream of the cauliflower mosaic virus 35S promoter (3...
متن کاملExpression of 35S::Pto globally activates defense-related genes in tomato plants.
The tomato (Lycopersicon esculentum) resistance gene Pto confers resistance to the bacterial pathogen Pseudomonas syringae pv tomato carrying the avirulent gene avrPto. Overexpressing Pto under the control of the cauliflower mosaic virus 35S promoter constitutively activates defense responses in the absence of pathogen infection and nonspecifically enhances disease resistance. To elucidate the ...
متن کاملCauliflower mosaic virus 35S promoter-controlled DNA copies of cowpea mosaic virus RNAs are infectious on plants.
Clones have been constructed that contain full-length cDNA copies of cowpea mosaic virus RNA1 and RNA2, downstream of the cauliflower mosaic virus 35S promoter. The clones, when linearized downstream of the viral sequences, give rise to cowpea mosaic virus-like symptoms when inoculated onto cowpea plants. Viral RNA and virions can be detected in the inoculated plants, demonstrating that the clo...
متن کاملTransformation of Cowpea Vigna Unguiculata with a Full-length Dna Copy of Cowpea Mosaic Virus M-rna
A full-length DNA copy of the M-RNA of cowpea mosaic virus (CPMV), supplied with either the 35S promoter from cauliflower mosaic virus (CaMV) or the nopaline synthase promoter from Agrobacterium tumefaciens, was introduced into the T-DNA region of a Ti-plasmid-derived gene vector and transferred to cowpea Vigna unguiculata cells. Southern blot analysis of the transformed callus tissue obtained ...
متن کاملCauliflower mosaic virus protein P6 is a suppressor of RNA silencing.
We infected a transgenic Arabidopsis line (GxA), containing an amplicon-silenced 35S : : GFP transgene, with cauliflower mosaic virus (CaMV), a plant pararetrovirus with a DNA genome. Systemically infected leaves showed strong GFP fluorescence and amplicon transcripts were detectable in Northern blots, indicating that silencing of GFP had been suppressed during CaMV-infection. Transgenic Arabid...
